Characterization of HLA genotype of cord blood units stock in Vietnam National Children’s hospital

Abstract
HLA (Human Leukocyte Antigen) gen is located on chromosome 6’s short arm including several loci. HLA genotyping is necessary for transplantation, especially in hematopoitic stem cell transplantation. A cross-sectional study to determine the frequency of HLA-A, HLA-B, HLA-BDR1 alleles of 233 cord blood units stock in Stem Cell Bank, National Children’s Hospital from year 2011-2014. Cord blood units were HLA genotyped by PCR-SSP method using AllSetTM Gold SSP Kit. The results showed that HLA-A locus has 17 alleles in which highest frequency alleles are A*11 (25.3%); A*02 (25.0%); A*33 (16.1%); A*24 (14.6%); A*29 (7.3%); A*26 (4.2%) and lowest frequency alleles are A*13; A*25; A*68; A*69; A*74 with 0.3%. HLA-B locus has 22 allleles in which highest frequency alleles are B*15 (28,4%); B*46 (9,8%); B*38 (8.0%); B*07 (7.6%); B*58 (6.9%); B*40 (6.2%) and lowest frequency alleles are B*18 (0.5%); B*37 (0.5%); B*08 (0.2%); B*14 (0.2%); B*67 (0.2%). HLA-DRB1 locus has 13 alleles in which highest frequency alleles are DRB1*12 (30.1%); DRB1*14 (11.4%); DRB1*09 (8.8%); DRB1*15 (8.5%); DRB1*04 (6.9%) and lowest frequency alleles are DRB1*16 (2.8%); DRB1*01 (0.5%). HLA-A, HLA-B and HLA-DRB1 loci has several haplotypes in which highest frequency haplotyes are A*11/B*15/DRB1*12 (17.5%); A*02/B*15/DRB1*12 (16.9%) and A*33/B*15/DRB1*12 (9.3%).
