Evaluation of the sub-chronic toxicity of Vietnam green coffee bean extract

Abstract
The study was conducted to evaluate the acute and subchronic toxicity of Vietnamese green coffee bean extract (VGCE) in experimental animals. The acute toxicity study was performed on Swiss mice via oral administration, using the fixed-dose procedure (OECD 420). The subchronic toxicity study adhered to the guidelines of the Ministry of Health. Wistar rats were orally administered VGCE at 300 mg/kg and 1500 mg/kg daily for four weeks. The results of the acute toxicity study showed that oral administration of VGCE up to 2000 mg/kg did not induce acute toxicity. The LD50 of VGCE by oral route in mice could not be determined due to the absence of mortality. In the subchronic toxicity study, no significant difference was observed in general condition, hematological parameters, liver and kidney function tests, or liver and kidney histopathology between the treated groups and the control group, or compared to baseline values before administration. In conclusion, a single oral dose of VGCE at 2000 mg/kg did not cause acute toxicity. Oral administration of VGCE at 300 mg/kg and 1500 mg/kg for four consecutive weeks did not result in subchronic toxicity in experimental animals.
