The activity assay of natural killer cells in normal adults

Abstract

To measure the levels of IF-γ cytokine in peripheral blood samples from healthy adults cultured in vitro with bacteria containing NK cell activators, across-sectional descriptive study was conducted on normal adult subjects with a sample size of 68 participants over 16 years old (including 36 men and 32 women), with ages ranging from 23 to 83 years old. The enzyme-linked immunosorbent technique (ELISA) was used to determine the activity of NK cells by measuring the amount of gamma interferon (IFγ) released in blood samples taken from peripheral veins and cultured in vitro at 37ºC in the presence of NK cell activators. The activity of peripheral blood NK cells in normal adults, expressed through the amount of IFγ released, was 1730.5 ± 833.19 pg/ml in men and 1734.21 ± 739.05 pg/ml in women, and 1732.03 ± 784.14 pg/ml in both. No clear trend in NK cell activity related to age was identified. There was no significant association between NK cell activity and total leukocyte count and peripheral blood lymphocyte count (p > 0.05). However, NK cell activity showed a strong positive correlation with the number of medium and large monocytes (p < 0.05, r = 0.938).
