Protective effects of S-Men in male mice with reproductive impairment induced by sodium valproate

Abstract
This study investigated the protective effects of S-Men against sodium valproate (NVP)-induced reproductive dysfunction in male mice. To induce reproductive impairment, mice were administered NVP at 500 mg/kg/day for five weeks, while concurrently receiving S-Men at two doses of 1096 mg/kg/day or 2192 mg/kg/day. The results indicated that S-Men significantly mitigated NVP-induced disorders, with the 2192 mg/kg/day dose exhibiting superior efficacy. Specifically, S-Men increased serum testosterone concentrations, enhanced sperm count and the percentage of morphologically normal sperm, and improved the weight of reproductive organs, particularly the levator ani-bulbocavernosus muscle complex. Furthermore, S-Men bolstered testicular antioxidant capacity by modulating MDA and GSH levels. Although testicular histological structures were not fully restored, functional reproductive indices showed marked improvement. In conclusion, S-Men exerts a potent protective effect and enhances reproductive function in NVP-challenged male mice, primarily by restoring the hypothalamic-pituitary-gonadal axis, improving sperm quality, and reducing testicular oxidative stress, with the most pronounced effects observed at 2192 mg/kg/day.
