The rs4747096 polymorphism of the ADAMTS-14 gene and its association with mri-assessed severity of knee osteoarthritis

Abstract
A cross-sectional descriptive study was conducted on 140 patients with knee osteoarthritis at Tien Giang General Hospital from June 2025 to January 2026 to characterize rs4747096 polymorphism of the ADAMTS-14 gene and investigate its association with the magnetic resonance imaging findings. The mean age of the participants was 51.99 ± 10.25 years old. The GG genotype demonstrated a higher proportion of severe osteoarthritis (66.7%) compared with AA (35.4%) and AG (40.0%). In multivariable analysis, after adjustment for potential risk factors, diabetes mellitus and the GG genotype of ADAMTS-14 were identified as factors associated with severe knee osteoarthritis, with odds ratios of 2.40 (95% CI: 1.12 - 6.21) and 1.92 (95% CI: 1.08 - 5.32), respectively. Collectively, these findings suggest that metabolic factors and genetic polymorphisms, particularly diabetes mellitus and the GG genotype of ADAMTS-14, are associated with severe knee osteoarthritis on magnetic resonance imaging.
