Vitexin, isovitexin contents and α-glucosidase inhibitory activity of mung bean seed coat extract

Abstract

Mung bean seed coat is an underutilized agro-industrial by-product with potential biological value. This study quantified vitexin and isovitexin in mung bean seed coat extract by HPLC-DAD, determined total phenolic and flavonoid contents, evaluated in vitro α-glucosidase inhibition, and used molecular docking to support the interpretation of interactions between major compounds and an α-glucosidase-related target. The material was collected in October 2025 from Nam Binh Import-Export Co., Ltd., Hung Yen province, where approximately 2 - 5 tons of seed coat by-product are generated monthly. VRE50 showed an extraction yield of 5.45% and contained 12.38% vitexin and 10.77% isovitexin in the extract. Total phenolic and flavonoid contents were 48.56 ± 1.46 mg GAE/g extract and 145.66 ± 4.37 mg QE/g extract, respectively. VRE50 inhibited α-glucosidase with an IC50 value of 148.36 ± 5.49 µg/mL. Molecular docking against 3TOP suggested potential interactions of vitexin and isovitexin with an α-glucosidase-related binding region. These findings provide preliminary evidence supporting further studies on mung bean seed coat as a vitexin- and isovitexin-rich by-product with in vitro α-glucosidase inhibitory potential.
