Association of the SLC5A2 rs9934336 polymorphism with fasting plasma glucose in patients with type 2 diabetes mellitus

Abstract

Given the critical role of genetic factors in glycemic control, this study investigated the association between the SLC5A2 rs9934336 polymorphism and fasting plasma glucose (FPG) levels in Vietnamese patients with type 2 diabetes mellitus (T2DM). A cross-sectional study was conducted on 123 diagnosed T2DM patients at Phu Nhuan District Hospital from July 2025 to November 2025. The SLC5A2 rs9934336 genotypes were identified using real-time polymerase chain reaction. FPG levels were compared across genotype groups, and multivariate linear regression was used to adjust for confounding factors. The frequencies of the GG, GA, and AA genotypes were 26.8%, 65.9%, and 7.3%, respectively. The GA+AA group exhibited significantly higher median FPG levels compared to the GG group (p = 0.013). In the multivariate regression analysis, each A allele was associated with a 10.3% increase in FPG levels after adjustment for confounders and False Discovery Rate (FDR) correction (p = 0.018). In conclusion, the SLC5A2 rs9934336 polymorphism is associated with FPG levels in T2DM patients, suggesting a potential role in glycemic regulation via the SGLT2 transporter.
