51. Experimental evaluation of acute and subchronic toxicity of Tinh Ky

Abstract

Tinh Ky soft pillformulation composed of  Rhizoma Polygonati (Hoàng tinh), Radix Astragali membranacei (Hoàng kỳ), Rhizoma Curculiginis (Sâm cau), and Cynomorium songaricum (Tỏa dương) is used in traditional medicine for the treatment of male infertility. This study was conducted to evaluate the acute and subchronic toxicity of Tinh Ky soft pills in experimental models.  The acute toxicity study was conducted on Swiss mice. The animals were administered increasing oral doses to determine the lowest dose causing 100% mortality and the highest dose causing no mortality. The subchronic toxicity study was conducted on Wistar rats, divided into three groups: the control group received distilled water at 2 ml/kg body weight. The treatment group 1 received Tinh Ky at 4.08 g/kg/day. The treatment group 2 received Tinh Ky 12.24 g/kg/day for 90 days. Body weight, general condition, heamatological and biochemical parameters, and histopathological changes in the liver and kidney were monitored thorough the study period . No lethal dose (LD50) could be determined for Tinh Ky soft pills, even at the maximum administrable oral dose of 51 g/kg body weight in mice. No subchronic toxicity was observed in Wistar rats administered 4.08 g/kg and 12.24 g/kg of Tinh Ky soft pills daily for 90 days.
